What is a regen?

When a player (Del Piero for example) retires, there is one less player in the database for the user to pick from...

Thus, the database then takes the retired Del Piero, and makes a brand new name, age, stats, description etc for him, and he is then placed in a random team.

This basically means that if there were 1000 players in the game to begin with, then by the end of the game, there will be 1000 players.

Hope that makes sense...

However please note...

That regens are in no way related to the player ho retires. If Beckham retires, the regen assigned to replace him could well be a Swiss defender, playing for Monaco with a PA of 10.

That's incorrect I believe. They call them newgens now, because the retired players are not "regenerated" anymore. It is totally random how many quality youngsters there are per year.
