First make sure you don't have the sound muted.
Next check the sound levels, goto control panel -> Sounds and audio devices, in the volume tab make sure the volume isn't muted or set too low, then click the advanced tab and make sure none of the volumes are muted or on a low level.
If they are all fine restart your computer.
If that doesn't work, assuming you are running Windows XP, goto the start menu -> run and type in services.msc click enter, in the window that opens should be a long list of services on your computer, scroll down to the one called Windows Audio, make sure it is started and on automatic, if its not double click it and on the window that opens, in the startup type select automatic, then click start further down, then click ok and exit services you should have audio back.
