NO you can't overwrite the original database, just save it as something else and use that database when starting a new game. Or use FMM and edit there PA's in the game when it's loaded.

I got a bit annoyed by that recently, every time I started a new game players I liked had dropped in PA, sometimes quite drastically from Astonishing 198 to Good-but-not-jaw-dropping 170.
So I edited the DB and gave my entire youth team a PA of 200, thinking I could manage them as well and Finally develop great youth players.
But I was actually disappointed with the results because, according to Genie Scout, the players wouldn't really turn out That Great...
I think a players CA also plays a big part in how they develop - if a player with a PA of 200 only starts with a CA of, say, 56 or something, then they end up just like your average Premier-League Standard players.
I think it would take a good deal of studying CA/PA and A LOT of trial and error to fully understand how it all all works. I've edited the DB loads of times (all in the name of fun and investigation) and have never really been satisfied with the results.
Perhaps it's best to leave it up to the Random PA generator, for best results.
But that's my opinion anyway.
